For more than 30 years, the Mantzoros Laboratory has been at the forefront of translational research, aiming to better understand, diagnose, and treat obesity and cardio-renal-liver-metabolic diseases, lipodystrophies and neuroendocrine disorders.
Current Projects:
- Translational research and new therapeutic approaches for metabolic diseases
- The effect of leptin, adiponectin, and other adipokines, as well as myokines, incretins, and IGFs in metabolic diseases.
- Creation of multi-omics machine learning models to uncover diagnostic and therapeutic insights in MASLD and cardio-renal-liver metabolic diseases
- Development of mendelian randomization and genetic studies in metabolic diseases, along with observational and interventional studies to explore the physiology and therapeutic potential of medications (e.g., proglucagon-derived peptides).
- Controlled phase I and phase IV clinical trials focused on obesity and its metabolic complications.
- Real-world studies to better understand the effects and side effects of pharmacotherapies in development for obesity and cardio-renal-liver-metabolic diseases.
- Participation in multi-center clinical trials on medications with dual agonism of glucagon and glucagon-like peptide-1 and their effect on metabolism and cardiovascular health.
- Development of investigator initiated randomized controlled trials to explore underlying mechanism and the therapeutic potential of nutrition, lifestyle interventions, and novel medications such as leptin or GLP-1 based receptor agonists or centrally acting anti-obesity medications.
About the Mantzoros Lab
Both the Human Nutrition Unit, led by Dr. Christos Mantzoros, and the Mantzoros Laboratory operate within the Department of Medicine at Beth Israel Deaconess Medical Center, a major teaching hospital of Harvard Medical School, in Boston, Massachusetts.
Dr. Mantzoros also directs the Endocrinology Section of the VA Boston Healthcare System (including the West Roxbury, Jamaica Plain/Boston, Bedford and Brockton campuses), affiliated with both Harvard and Boston University School of Medicine. An additional affiliation with the University of Massachusetts School of Medicine is in progress.

Colleagues and collaborators at Harvard and other institutions around the world include many leaders in the fields of molecular medicine, metabolism and clinical epidemiology, creating a rich environment for research and education.
Our academic team consists of practicing physicians who are active in performing basic and clinical research, providing clinical care, teaching at the undergraduate and graduate levels, and providing hands-on training to medical students, residents, and fellows in Internal Medicine, with a focus on Endocrinology, Diabetes, and Metabolism and more specifically Obesity and Cardio-Renal-Liver-Metabolic Disorders.

Dr. Christos S. Mantzoros
The Director of the Unit, Dr. Christos Mantzoros, has been Board Certified in Internal Medicine, Endocrinology, Diabetes and Metabolism as well as Clinical Nutrition. He serves as the Chief of the Endocrinology Section at the VA Boston Healthcare System and as the Director of Human Nutrition at Beth Israel Deaconess Medical Center and the Joslin Diabetes Center.
Dr. Mantzoros is a Professor of Medicine at Harvard Medical School and an adjunct Professor of Medicine at Boston University School of Medicine. He is also active with the Scholars in Clinical Science Program at Harvard Medical School, a postgraduate Master's level two-year-long program that provides formal training in Clinical and Translational Research for medical doctors at Harvard Medical School-affiliated institutions; as well as with the International Research Initiative, a one year training program designed for physicians who want to pursue rigorous clinical or basic science research careers.
Dr. Mantzoros has served on the advisory boards of foundations and academic institutions as well as several pharmaceutical and diagnostic companies. He is on the Board of ALBA Graduate Business School.
Dr. Mantzoros also serves as the Editor in Chief of the journal Metabolism, Clinical and Experimental (Impact factor: 13.94, Cite score: 20.5).

What we do
Pioneering work on leptin, adiponectin, and IGFs has elucidated the physiology of energy homeostasis and led to FDA-approved therapies and groundbreaking contributions to pharmaceutical innovation. We conduct multi-omics and machine learning studies to uncover actionable diagnostic and therapeutic insights into steatotic liver disease, Mendelian randomization to identify causal relationships and raise hypotheses, animal preclinical studies, observational and interventional physiology studies to explore the therapeutic potential of proglucagon-derived peptides, randomized controlled clinical trials to examine the impact of novel medications in the pipeline against obesity and metabolic disorders, population-based studies to elucidate the impact of nutrition on metabolic health and real-world studies to examine the impact of anti-obesity and antidiabetic medications on cardiometabolic and neurocognitive disorders, psychiatric outcomes, fatty liver disease, and malignancies.
